Buckley, who is Irish, had swept the awards circuit leading up to the ceremony, winning in her category at the Critics Choice Awards, Golden Globes, BAFTAs and Actor Awards. She is the first Irish performer to win a best actress Oscar.
A front-runner in the category, Buckley was nominated alongside first-time nominees Rose Byrne for If I Had Legs I´d Kick You' and Renate Reinsve for Sentimental Value.
Two-time winner Emma Stone was also nominated for Bugonia, as was two-time nominee Kate Hudson for Song Sung Blue.
Hamnet, an adaptation of Maggie O´Farrell´s prizewinning historical fiction novel, dramatises the mysterious private life of the famed playwright and poet Shakespeare and his wife Anne Hathaway - who was also called Agnes.
The plot centers on the death of the couple´s 11-year-old son, Hamnet, and the intense grief that followed.
Jordan won the best actor Oscar for playing twins confronted with pure evil in vampire race fable Sinners - tortured fighters typical of the roles director Ryan Coogler has repeatedly created for him.
The actor made good on the momentum he gained by winning the SAG Actor Award two weeks ago to bring home an Academy Award in his first try.
He bested 'Marty Supreme star Chalamet, who had been the frontrunner for most of Hollywood's awards season, along with Leonardo DiCaprio of One Battle After Another, Wagner Moura for The Secret Agent, and Ethan Hawke for Blue Moon.
At age 39, Jordan joins a small circle of Black actors who have won the prestigious best actor Oscar, after Sidney Poitier, Denzel Washington, Forest Whitaker and Will Smith.
'I stand here because of the people who came before me,' an emotional Jordan told the audience on Sunday evening.
Sinners, a supernatural tale of racial segregation in 1930s Mississippi, was a box office success in large part due to Jordan's compelling performances as Smoke and Stack, World War I veterans who return home after working in organized crime in Chicago.
